Frizzled-8是WNT蛋白10個(gè)哺乳類卷曲受體之一。卷曲蛋白細(xì)胞外的半胱氨酸富集區(qū)與WNTs的密切關(guān)系可達(dá)到nM級別。當(dāng)被作為重組蛋白時(shí)，F(xiàn)rizzled-8的細(xì)胞外區(qū)域就成為可溶的受體，以阻止細(xì)胞中WNTs與Frizzled的結(jié)合。
StemRD公司的Frizzled-8細(xì)胞外主蛋白是與人IgG1細(xì)胞Fc片段的N-終端相融合的。這種融合保證了體內(nèi)外蛋白的穩(wěn)定性，以及重組蛋白二聚體對WNTs的高親和性。研究基因科技的科學(xué)家曾使用與之類似的蛋白Frizzled-8 Fc融合，證實(shí)了動(dòng)物模型中WNT封鎖物的抗腫瘤效果（DeAlmeida VI et al，Cancer Res.2007,67:5371）。

Frizzled-8 is one of the 10 mammalian Frizzled receptors for WNT proteins. The extracellular cycteine rich domain of Frizzleds bind to WNTs with nM affinity. When supplied as recombinant protein, this extracellular domain of Frizzled-8 acts as a soluble receptor, blocking the binding of WNTs to Frizzled on the cells.
StemRD's Frizzled-8 extracellular domain protein is fused to the Nterminus of the Fc domain of human IgG1. This fusion allows stabilization of the protein in vitro and in vivo and dimerization of the recombinant protein rendering high-affinity binding to WNTs. The Frizzled-8 Fc fusion has been used by Genentech scientists to show anti-tumor effect of WNT blockade in animal models (DeAlmeida VI et al, Cancer Res. 2007, 67:5371).
Expression is from CHO cells cultured in serum-free and protein-free medium. Purification of the fusion protein from conditioned culture medium is performed using protein A chromatography.
